The human papilloma virus (HPV) family of DNA tumor viruses includes HPV16, a 'high-risk' sexually-transmitted HPV that can lead to cervical, anal, vulvar, head, neck, and penile cancer. The HPV16E1/E4 protein is expressed abundantly in cells supporting viral DNA amplification, but is lost during malignant progression. HPV16E1/E4 causes G2 cell cycle arrest by associating with and preventing the nuclear entry of Cdk1/cyclin B1 complexes. HPV16E1/E4 also interacts with cyclin A and Cdk2 during the G2 phase of the cell cycle, and this association may increase the efficiency with which HPV16E1/E4 is able to prevent mitotic entry. HPV16E1/E4 also associates with keratin intermediate filaments and causes the network to collapse. The protein encoded by this gene is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is found in the asymmetrical unit membrane (AUM) where it can complex with other transmembrane 4 superfamily proteins. It may play a role in normal bladder epithelial physiology, possibly in regulating membrane permeability of superficial umbrella cells or in stabilizing the apical membrane through AUM/cytoskeletal interactions. Recognizes a protein of 34 kDa, identified as a subunit of Replication Protein A (RPA) (also known as human single-stranded DNA binding protein, or HSSB). The finding that mutations in DNA mismatch repair genes are associated with hereditary nonpolyposis colorectal cancer (HNPCC) has resulted in considerable interest in the understanding of the mechanism of DNA mismatch repair. Initially, inherited mutations in the MSH2 and MLH1 homologs of the bacterial DNA mismatch repair genes mutS and mutL were demonstrated at high frequency in HNPCC and were shown to be associated with microsatellite instability. A member of the mismatch repair family, GTBP (also designated MSH6), is an MSH2-related protein that binds to DNA containing G/T mismatches. Findings suggest that the mismatch-binding factor in human cells is composed of a heterodimer of GTBP and MSH2. Complement C1q A Chain (C1qa) is a subunit of C1q. C1q is composed of 6 polypeptide chains each of C1qa, C1qb, and C1qc. C1q is a component of the classical complement pathway, and thereby plays an important role in the immune response. Each C1q subunit contains an N-terminal collagen-like region and a C-terminal C1q globular domain. The presence of receptors for C1q on effector cells modulates its activity, which may be antibody-dependent or independent. Macrophages are the primary source of C1q, while anti-inflammatory drugs as well as cytokines differentially regulate expression of the mRNA as well as the protein. PAI-RBP1 (plasminogen activator inhibitor 1 RNA-binding protein), also known as SERBP1 (SERPINE1 mRNA-binding protein 1), CGI-55, CHD3IP (chromodomain helicase DNA binding protein 3 interacting protein), HABP4L or PAI-RBP1, is a membrane-associated protein that localizes to the nucleus, the perinuclear region of the cytoplasm and the plasma membrane. PAI-RBP1 is believed to play a role in the regulation of mRNA stability, as it specifically binds to the CRS (cyclic nucleotide-responsive sequence) motif of the PAI-1 mRNA and acts to stabilize the mRNA and regulate its expression. In addition, PAI-RBP1 interacts with Mi2- and may be involved in chromatin remodeling. PAI-RBP1 also interacts with PGRMC1 and participates in the transduction of Progesterone s antiapoptotic action in ovarian cell types.
